Plasmid Handling and Storage

Short-term Storage

Bacterial cultures in LB medium (e.g., an overnight liquid culture) should be stored at 4 °C upon delivery. Under these conditions, the culture generally remains viable for up to one week.

Long-term Storage

For long-term storage, plasmid DNA is typically stored at –20 °C, where it remains stable for years. For preservation of bacterial strains, glycerol stocks are prepared by mixing cultures with 25% glycerol and storing them at –80 °C, ensuring bacterial viability for months to years.
